Drew Cook attended Regina High School and lived in Iowa City, Iowa. He was named Class 1A first-team all-state as a junior and senior, and was the MVP of the Class 1A state championship game as a junior and senior. He was named Class 1A Player of the Year by Cedar Rapids Gazette and Offensive Player of the Year by Iowa City Press-Citizen as a senior.

He redshirted in 2015, and moved from quarterback to tight end and saw limited action. He was twice named Academic All-Big Ten.

His father is Marv Cook, who played TE at Iowa in the late 1980's and played many years in the NFL.
